This Lenti-X qRT-PCR Titration Kit and a real-time PCR instrument can be used to rapidly quantify the virion content of HIV-1-based lentiviral supernatants. The viral genome copy number in a supernatant sample is determined by first purifying the viral RNA, and then comparing the RT-PCR threshold cycle (Ct) of an appropriate dilution of purified viral RNA to sample curve Ct values that are derived from a calibrated, serially diluted control viral RNA template.

By quantifying the amount of lentivirus inserted into the target cell's nuclear DNA, it is possible to more accurately measure the actual titer (effective titer) and MOI (Multiplicity of infection) present within the Lentiviral supernatant stock. Through this, it is possible to predict how many viral genomes have been inserted into the target cell.

The provirus quantitation kit is a product that can rapidly quantify the copy number of integrated lentivirus (provirus) inserted within transduced cells. This product includes a genomic DNA purification kit, 200 doses of TB Green qPCR premix, and a calibrated control template.

The primer included in this product is designed to amplify the HIV-1 genome region adjacent to the Packaging Signal. This primer is compatible with all self-developed lentivirus vectors as well as commonly used HIV-1-based lentiviral vectors.
